Antibody-drug conjugates (ADCs) and bispecific antibodies are transforming the treatment landscape for solid tumors by providing targeted options for patients with limited alternatives. In lung cancer, bispecific agents targeting PD-1 and VEGF demonstrate significant efficacy in PD-L1 negative populations, addressing a critical clinical need. Breast cancer management has evolved with the integration of trastuzumab deruxtecan, which now outperforms traditional therapies, prompting new strategies for sequencing and combination regimens to manage residual disease. Endometrial cancer treatment has seen a paradigm shift due to HER2-targeted ADCs, which yield unprecedented response rates in recurrent disease and have secured accelerated FDA approvals. Furthermore, B7H3-targeted ADCs are emerging as viable therapies for metastatic castration-resistant prostate cancer, maintaining efficacy even in patients previously treated with PSMA-directed radiopharmaceuticals. These advancements underscore the potential of novel payloads and target-specific therapies to improve outcomes across diverse oncological indications.
